3,4,5-trimethoxybenzoate Methyl ester 3,4, 5-Trimethoxybenzoate is synthesized from Gallic acid and is mainly used to produce trimethoprim (TMP), sulfonamide synergist intermediates, and many other drugs.
It is prepared by the methylation of gallic acid and dimethyl sulfate. Add water, gallic acid and dimethyl sulfate to the reaction pot, drop sodiumhydroxide solution at 15-35ºC, and stir at about 40ºC for half an hour. A second batch of dimethyl sulfate was added and sodiumhydroxide solution was added at about 40ºC to a pH of 8-9. After 1h reaction, it was cooled, filtered, washed and dried to get methyl trimethoxy benzoate with yield above 90%. The methylation and esterification reactions mentioned above can also be carried out in water and chloro form using tetrabutyl ammonium bromide as phase transfer catalyst. The product can also be prepared by using tannic acid as raw material or by esterification of 3,4, 5-trimethoxy benzoic acid and meth anol.
